mRNA and miRNA Expression Profiling

  BMGen offers mRNA and miRNA expression profiling services on Affymetrix, Agilent and Illumina microarray platforms. These microarray platforms have both the genome-wide and custom applications for a number of different organisms.

Affymetrix high-density 25-mer oligo arrays: GeneChip technology platform consists of high-density oligo microarrays and is ideal for fast expression screening of thousands of genes. This technique is based on a hybridization of biotin-labelled sample on a microarray. Binding targets are detected by streptavidin-conjugated fluorochromes. The resulting intensity data is analyzed by Gene Expression Console Software. Affymetrix GeneChips for gene expression analysis are available for around 30 different species including human, rodents, insects, plants and bacteria.

Agilent 1-color and 2-color 60-mer oligo arrays: The Agilent 60-mer oligonucleotide microarray platform can be used on both the one- and two-color gene expression applications. The principle of this technique (two-color) is to compare the relative abundance of expressed sequences in two RNA samples (test and reference) labeled with two different fluorescent colors (Cy3 and Cy5). The resulting ratio data is analyzed by Feature Extraction software. The other option is to hybridize one sample per array (one-color, labeled with Cy3) and compare the expression of different samples computationally. The whole genome spanning catalogue arrays are printed as multi-pack 4 x 44k or 8 x 15k formats for a number of different organisms. Alternatively, custom arrays are available as 1 x 244K, 2 x 105K, 4 x 44K, or 8 x 15K format.

Illumina 50-mer oligo arrays: Illumina gene expression microarrays allow rapid and cost-effective whole-genome expression profiling for mouse, human and rat. Direct hybridization assay includes a first- and second-strand reverse transcription step, followed by in vitro transcription (IVT) that incorporates biotin-labeled nucleotides to the nascent cRNA strand. Subsequent steps include array hybridization, washing, blocking, and streptavidin-Cy3 staining. The gene expression analysis of fragmented RNA is also possible using DASL assay. The array pre-processing is carried out using GenomeStudio software.
